UPLC Fingerprinting Study of Dipsacus asper Wall. and Its Formulated Granules
Objective: To establish the UPLC fingerprints of Dipsacus asper W. and formulated granules, and to provide an effective method for the quality control of the formulated granules of Dipsacus asper W. Methods: UPLC was used to simultaneously determine 10 batches of Dipsacus asper W. and 10 batches of the formulated granules of Dipsacus asper W. and to develop the fingerprint profiles. The chromatographic column was Agilent ZORBAX Eclipse plus C18 (2.1 mm × 100 mm, 1.8 μm); the flow rate was 0.2 mL∙min −1; the detection wavelength was 212 nm; the column temperature was 30˚C; the injection volume was 1 μL; and the mobile phase was a binary gradient elution of 0.1% phosphoric acid in water (A)-acetonitrile (C). Result: There were 25 common chromatographic peaks in 10 batches of Dipsacus asper W., 32 common chromatographic peaks in 10 batches of Formula granules, and 25 common peaks were determined between 10 batches of Dipsacus asper W. and 10 batches of formula particles, with high separation between each peak. The similarity between 10 batches of Dipsacus asper W. and 10 batches of formula particles is greater than 0.8, and the similarity between 10 batches of formula particles is greater than 0.9. Conclusion: The fingerprints of Dipsacus asper W. and Formula granules were stable and reliable and the number of chromatographic peaks of the herbs and the formulated particles had a large difference, indicating that the chemical compositions of the herbs had changed and differed after the preparation process of extraction, concentration and granulation.
Dipsacus asper Wall.
续断(Dipsacus asper Wall.),又名和尚头,为川续断科川续断的干燥根,多年生草本植物,因能“续折接骨”而得名。其味苦、辛,性微温,归肝、肾经,能够补肝肾、强筋骨、续折伤、止崩漏。在续断中发现的如三萜皂苷类、环烯醚萜苷类、生物碱类及挥发油等有效成分
随着社会的发展和科技的进步,传统的以煎、煮药物的提取方式已不适用于现代化的发展,服药不便、质量标准不易掌控、搬运储存困难等缺点严重影响了中药的发展
近年来,中药配方颗粒的市场迅速扩大,其质量问题也受到越来越多的关注。中药配方颗粒存在的质量标准不统一、生产工艺水平参差不齐、国家监管难度大等问题难以解决
因此,本研究通过UPLC技术对续断及其配方颗粒进行分析,建立续断及配方颗粒的UPLC指纹图谱,建立完善可行的质量评价方法,为确保续断及其配方颗粒的一致性评价及质量控制提供理论依据。

取10批续断药材,粉碎,过4号筛,各取约1.0 g,精密称定,置具塞锥形瓶中,加甲醇25 mL,称定质量,超声提取(功率250 W,频率35 k Hz) 30 min,放冷,再称定质量,用甲醇补足减失的质量,摇匀、过滤,取续滤液过0.22微孔滤膜,即得续断药材供试品溶液;取10批续断配方颗粒,研细,各取约1.0 g,精密称定,置具塞锥形瓶中,按药材供试品溶液制备方法,制备续断配方颗粒供试品溶液。
取同一批次续断药材样品,按“溶液的制备”项下制备供试品溶液,按“液相指纹图谱的色谱条件”项下色谱方法连续进样6次,并记录各共有峰的保留时间和峰面积,计算各共有峰的相对保留时间和相对峰面积的RSD均小于3.0%,表明仪器精密度良好。
取同一批次续断药材样品,按“溶液的制备”项下制备供试品溶液,按“液相指纹图谱的色谱条件”项下色谱方法分别于0、2、4、8、12和24 h进样测定,并记录各共有峰的保留时间和峰面积,计算各共有峰的相对保留时间和相对峰面积的RSD均小于3.0%,表明样品溶液在24 h内稳定。
取同一批次续断药材样品6份,按“溶液的制备”项下制备6份供试品溶液,按“液相指纹图谱的色谱条件”项下色谱方法分别进样,并记录各共有峰的保留时间和峰面积,计算各共有峰的相对保留时间和相对峰面积的RSD均小于3.0%,表明该方法重复性良好。
按确定的方法制备分析样品,按确定的色谱条件分别对10批续断药材、10批续断配方颗粒进行测定,得到各批次样品UPLC色谱图,分别导入中药指纹图谱相似度评价系统(2012A),得到续断药材、续断配方颗粒、续断药材及配方颗粒色谱指纹图谱及对照色谱指纹图谱(
以0.1%磷酸水和乙腈进行梯度洗脱,峰形较好,分离效果理想,建立的续断药材及其配方颗粒的指纹图谱中,10批续断药材确定25个共有色谱峰,10批续断配方颗粒确定32个共有色谱峰,10批续断药材与10批配方颗粒确定25个共有峰。以5号峰为药材和配方颗粒的参照峰,计算各共有峰的相对保留时间与相对峰面积,见

本文初步建立了续断药材和配方颗粒的UPLC指纹图谱,对10批续断药材和10批续断的配方颗粒进行了研究,发现药材与配方颗粒剂之间的共有峰存在差异,说明药材经提取、浓缩、制粒等制备工艺后,化学成分有所变化,并采用指纹图谱相似度评价软件对续断药材和配方颗粒进行了相似度评价,续断药材和配方颗粒均有较好的相似度,相关系数均大于0.8。10批药材中S2相似度最低,成分存在差异,说明不同药材之间成分存在个体差异,采收时间、生长年限和土壤类型等因素可能是造成差异的主要原因,因此在制备成配方颗粒时需要注意对药材进行分析鉴别,保障续断来源的可靠性,确保配方颗粒的有效性和准确性。
配方颗粒中的共有峰较药材中的共有峰多,可能是由于制剂工艺过程中(如高温提取、浓缩、辅料添加等)产生的,这些成分是在制剂过程中其他成分受热相互作用产生还是由其他成分转化,有待进一步的研究
